Plans for the world’s first clinical trial for a vaccine targeting Alzheimer’s disease-like characteristics in those with Down’s syndrome have been announced.
The study will test AC Immune’s vaccine ACI-24 and is being conducted in collaboration with the University of California, San Diego (UC San Diego) Down’s Syndrome Research and Treatment Center. Funding is provided by a significant grant from the US National Institutes of Health (NIH) and an additional grant from the LuMind Research Down’s Syndrome Foundation. This is the first public/private collaboration for a clinical trial in the field of Down’s syndrome.
Individuals with Down’s syndrome (DS) have an extra copy of chromosome 21 which carries the gene for APP encoding the precursor protein of Abeta, one of the hallmarks of Alzheimer’s disease (AD). An important consequence is that individuals with DS develop AD-like characteristics at a rate three to five times greater than that of the general population and at a much younger age. Further, AD-like characteristics develop in more than 98% of people with DS over age 40 with up to 80% developing associated dementia over the age of 60. It is estimated that there are 6 million people with DS worldwide, with 400,000 in the United States.
Trial participants will be adults with DS. The objectives of the trial include studying safety and tolerability of ACI-24, its effect on induction of antibodies against Abeta, clinical and cognitive measures in adults with DS and its effect on biomarkers of AD-like pathology in DS. Participants in the study will be treated for 12 months, with 12 months follow up.

Additionally, this ground-breaking clinical trial could enhance our understanding of early intervention and prevention of Alzheimer’s in general.”

That both public and private funding sources support the study signifies the importance attached to Alzheimer’s disease and the valuable insights that will come from studies of this disorder in Down’s syndrome. We wish to thank our colleagues as we eagerly look forward to helping people with Down’s syndrome and their families and loved-ones.”

I am other of a son with DS and ad parents we have a vested interest in this research and it’s progress for over a year and have eagerly anticipated this phase! I would greatly appreciate it if someone could contact me to discuss the possibilities of including our son in this trial or having him as a qualified candidate for future research. I am not sure if we would qualify as we are in Canada but if it’s any consolation it’s been pre discussed with his geneticist and I have the support of our medical team. I eagerly anticipate your response!

This is a promising development. As a parent of a 14 year old daughter with DS I am most appreciative of the work of researchers and LuMind. She participated in the Clematis drug study and it was a great experience.
Our son, age 36, has early onset dementia. He is on Namenda and Excelon. I am interested in discussing this with other parents going through the same thing.
My daughter is 8 months old, with DS. Have read up a bit on the dementia that could occur in my child. Feel this is a good study to be carried out. Love and support.x

I take care of my forty-seven year old brother who is a fairly high functioning and has Downs. I would be extremely interested in participating in a trial. We live in El Paso, Texas. He has days when we have the same conversation a few times, and days when he is right on. There are friends he may not recognize and strangers he is ready to hug. My son with Down Syndrome he is 25 was diagnosed with Dementia a year ago. He started to stop going out into the community and doing the things he liked doing such as swimming and socializing with friends. He also has a hard time taking a shower, shaving etc.. He goes to his Post High program has hard time getting up and getting on the bus. He never use to hit us before however we are now getting hit. He seems confused often .. I have never meet another person with DS this young that has Dementia.
